Test Sampling Condition of Lilium Seed Germination with Automatic Counting Machine

Lily has a variety of classifications, and its seeds are also in two forms, cotyledon unearthed and cotyledon clay. The clay-retaining lily seeds have relatively large particles, slower germination and dormancy, whereas the unearthed ones are just the opposite. Because of the different habitats and species of lilies, the main factors influencing seed germination are also different, and species such as Lilium longifolia and Lilium persicae are growing. In the environment with less light, air and soil moisture, the seed germination rate is almost not affected by light, but the temperature has a significant effect on it. On the basis of previous work, the effects of light intensity and temperature on the germination of Lilium zizanioides seedlings under different habitat conditions, different location effects and different storage time were comprehensively discussed, providing basic data for the protection and utilization of Lilium germplasm resources. Sample counting can be performed using an automated pelletizer during the test.

Take a clean petri dish, put a filter paper in it, put the seeds rinsed with tap water on the filter paper, and then add tap water to the petri dish so that the seed can float on the filter paper, so that the seed coat can absorb water and swell. After 4 hours, the excess water was sucked with a suction pipe, and the water-absorbing seeds were placed neatly on the filter paper, using filter paper and tap water as a matrix.

In the experiment of germination of Lilium lucidum seeds stored at room temperature for one year by automated counting instrument, it was found that the seed germination rate was slightly different at 27Lmo1m-2s1, 35Lmo1m-2s' and 20e and 25e under light at 20e constant temperature. More than the new seeds collected in that year, about 2% to 4% higher; in 30e temperature and light, the germination rate of Lilium sylvestris seeds stored for one year is higher than that of the seeds of the current year, and the complete germination time is also reduced than the current year seed. For 5 days, the seed germination rate of Chen seeds was significantly lower than that of new seeds in the dark at 5e, 10e, and 15e. The seed germination rate of seeds after one year of storage did not decrease in the above treatments. Similar phenomena were also found in other species of the genus Lilium. The reason for this has not yet been found to be reasonable.
